چکیده

Introduction Peptide mapping is a common technique in the biopharmaceutical industry to characterize monoclonal antibodies (mAbs) for the determination of product identity and stability. Many conventional sample preparation methods are time consuming with digestion times of several hours and can introduce modifications such as deamidation, oxidation, and carbamylation.1 In this study, two classic in-solution digestion approaches were compared to the recently developed SMART Digest kit method to quantify the extent of posttranslational and chemical modifications of a therapeutic recombinant mAb. Proteolysis in mixed organic-aqueous solvent systems: applications for peptide mass mapping using mass spectrometry.

The rate of protein digestion imposes significant limitations on high-throughput protein identification using mass spectrometry. In this report, we demonstrate that proteins are readily digested by trypsin in the presence of organic solvents such as methanol, acetone, 2-propanol, and acetonitrile. The rates of protein digestion in organic solvents, as indicated by the abundances of digest fragm...

Modeling of Partial Digest Problem as a Network flows problem

Restriction Site Mapping is one of the interesting tasks in Computational Biology. A DNA strand can be thought of as a string on the letters A, T, C, and G. When a particular restriction enzyme is added to a DNA solution, the DNA is cut at particular restriction sites. The goal of the restriction site mapping is to determine the location of every site for a given enzyme. In partial digest metho...

Thermal denaturation: a useful technique in peptide mass mapping.

The use of thermal denaturation of proteins prior to in-solution digestion and mass spectral peptide mass mapping is reported. Thermal denaturation is preferred over chemical denaturation because it does not require purification/concentration prior to mass spectral analysis. Enzymatic digestions of proteins that are resistant to proteolysis are significantly enhanced by thermal denaturation. Na...

Radioiodine D amino acids labeling of Rituximab, a new method for enhancing the radiopharmaceutical targetingand biostability

Introduction: Radioimmunotherapy (RIT) is a very promising new therapy for the treatment of recurrent B-Cell non-Hodgkin's lymphoma (NHL). Iodine-131 is the most frequently used nuclide in clinical RIT, but its usefulness has been limited by dehalogenation of monoclonal antibodies labeled via conventional methods. To circumvent this problem, we have synthesized a tr...

Evaluation of Spent Catalyst Structure Changes during Ethylbenzene Dehydrogenation in Different Technologies

Ethylbenzene (EB) dehydrogenation to styrene (SM) on an industrial scale is generally performed using classic and SMART (Styrene Monomer Advanced Reheat Technology) technologies. In the current study, spent catalysts structural changes through classic and SMART technologies were investigated and compared with the fresh catalyst. For this purpose, XRF, XRD, SEM-EDX, FT-IR, BET and crushing s...
